You can do that and no one is stopping you.
Staying for 2 years won't show that you are persistent.

Yep. Staying for 2 years before applying will just be showing them that you know how to follow rules. That is something Singapore likes. Oppies are persistent, governments don't like them.
On second thought, go ahead and make a nuisance of yourself. It might be a good learning experience.

Someone shared that he has applied 8 times in 8 years, all got rejected. Persistence is not demonstrated through repeated applications.

Think you have to take into account who you work for. Is your employer's business doing well? Doesnt necessarily have to be about the size of the company, but more so if the company is profitable (ie paying tax). I say this because I have 2 other friends that submitted the same time as me in similar industries, their PR got approved but mine rejected. And based on my deductions, it is boiled down to the difference in employer.
